Deathtouch

 {Vampire Nighthawk} and  {Polukranos, World Eater} you activate Poly's monstrosity and deals the damage to vamp. As a requirement vamp deals damage equal to it's strength back. Does the deathtouch apply since it was combat damage?

Deathtouch applies to any damage. Monster Hydra will die

To clarify, it's not Combat Damage.  Combat damage is only damage dealt during a Combat Damage Step.  However, Lifelink and Deathtouch (and Infect and Wither) apply to ANY damage the creature itself deals, whether it's through Combat, Fighting, Polukranos' Ability, {Sou's Fire}, etc.

Yes. The creature is dealing the damage, not burning anger.
