Does a 1/1 with deathtouch kill a 6/5 with hexproof?

Yes it does.  Hexproof will only prevent targetting of an opponents ability.  Deathtouch is a property of damage and can go through hexproof

to be more specific, deathtouch makes any amount of damage lethal, so a 0/4 defender that gains deathtouch won't do lethal damage.

That's actually incorrect. A creature must be able to assign damage. See rule:

"119.8
If as source would deal 0 damage, it does not deal damage at all. That means abilities that trigger on damage being dealt won't trigger...."
