If you have a static effect already on the field, say "All creatures you control get +2/+2." then yes. But you wont be able to cast an instant like {Mutagenic Growth} to save it.

Well, what you could do is, while the Shadow is on the stack, respond by casting the growth targeting something else in play. You can't target the shadow, because it's still a spell. Thats one way to reduce your life by two.

{Death's Shadow} doesn't place  -1/-1 counters on its self. It's giving its self -x/-x which is like {Dismember} type ability. So {Merlina, Sylvok Outcast} won't work this time.
