At my local prerelease there has been a couple issues with the understanding of how the card  {Silence} can be played. What was ruled at the prerelease has already happened and I will not go over that because I was not directly involved with the conflict, but I want to know one thing:

Can you or can you not play  {Silence} during your opponent's upkeep to prevent them from having a chance to play spells during their mainphases/rest of their turn? If not, when is the earliest opportunity to cast an instant on your opponent's turn?

Yes you can cast during the upkeep.

This was judged at my tournament as well; I was the one playing Silence.
I could cast during opponent's upkeep (starting a stack), opponent therefore gets only that one opportunity to cast an instant of their own in response on the same stack (in his case {Volcanic Geyser}). His instant resolves first, then mine and he is shut down thereafter.
