So if my opponent plays a spell then i use {Cancel} the countered spell goes to the graveyard right?

Yes.  {Cancel} goes on the stack last and resolves

Its my opponents turn, he taps a card to use an ability, after it is tapped, i then tap royal assassin to destroy that creature. Can that ability still be used or does my action of destruction resolve first??

I believe the abilities go on the stack in the order in which they are activated so if you tap your {royal assassin} once he taps his creature the royal assassin ability resolves first since it is on the top of the stack. Don't take my word for it though. Wait until black jester answers this one. But if I would have to bet on it I would bet I am right

Thanks for the intro! You are right, both abilities will go on the stack, first their tap ability, then your {Royal Assassin}'s ability on second.

Let's say the creature in question is {Gideon's Lawkeeper}. It's your opponent's turn and they activate his "Tap target creature." ability, targeting your {Grave Titan}. The ability goes on the stack.

Then, you respond by activating the assassin's ability, targeting the Lawkeeper. This gets put on the stack, on top o the other one. Now, if nobody has any other plays, the stack will "resolve" starting at the top. Last in, first out. So the last thing put on the stack will be the first to go off.

The Lawkeeper will die. Abilities on the stack are separate from what created them, so the "tap" ability will still resolve, tapping the Titan.
