if they  {Duress} My instant can I cast the instant after he targets.

No. You can cast instants before Duress begins resolving, but once a spell starts to resolve you can't do anything the card doesn't say.

No. Once the spell starts resolving, it's too late. You gotta cast it in response to the duress before you reveal and he chooses for you to discard.
