So if my opponent has a creature enchanted with {invisibility} and I play {polymorphist's jest}, can I block his creature?

Nope. The enchantment is still attached and says "enchanted creature cant be blocked except by walls." If the enchantment gave the ability to the creature it would qork, but that is not the case.

The jest would strip a {Phantom Warrior} or {Triton Shorestalker} of their unblock ability but since here is another aura granting it then it sticks
