If {Doran the Siege Tower} was in combat with a first strike creature and was dealt two damage would he then assign 3 damage or still 5

I don't understand this. I thought if you did like 3 damage, you could finish it off with {Shock} or something. Why is this any different?
This is true. When something with 5 toughness is dealt 3 damage, it still has 5 toughness. However, until the end of the turn the game remembers that it had 3 damage dealt to it (3 damage is "marked") in case enough more is dealt to kill it. In this case, 5 toughness - 3 marked = 2 more damage to finish it off.

It's not. The toughness is still 5, it just has 3 damage on it. Adding a {Shock} will make it 5 damage marked, equal to its Toughness, which is enough to kill it.
