When a permanent, for this example, a creature would die from a source such as, let's say, Doom Blade, and I Regenerate it, but then it gets targeted again with another Doom Blade, could I Regenrate it again?
Since it got tapped from the first Regenerate, could I do it again?
Yes, as long as you can pay the activation cost.

The word "instead" doesn't appear on the card but is implicit in the definition of regeneration. "Regenerate [permanent]" means "The next time [permanent] would be destroyed this turn, instead remove all damage marked on it and tap it. If it's an attacking or blocking creature, remove it from combat." Abilities that trigger from damage being dealt still trigger even if the permanent regenerates. See rule 701.12.
It says to tap it as an effect not as a cost.
