Title: Dodging assassinate

I my opponent plays  {Assassinate} and I respond with a {burst of strength} does his spell fizzle because my creature is no longer tapped and therefore an invalid target?

Would that work against a creature effect as well such as the effect on royal assassin?

Sure! An effect has to follow the rules, whether the source is a creature, sorcery, artifact, or whatever.
