Say I have a {Knight of Glory} and my friend has a {Butcher Ghoul} with {Predator's Gambit} can I block it with the knight of glory since it has protection from black?

Protection will not have any effect here. With intimidate, the Ghoul can only be blocked by artifact or black creatures. The knight is neither, and will not be able to block.

In your favor his creature won't be able to block yours when it attacks either.
