My brother has OLD cards that have abilities that are not in my MtG rulebook.
First off, what is the card type "Mana Source?"
Can I destroy it with a "Destroy target land?"
Also, he has cards that "Destroy ALL lands" and "Destroy ALL creatures." Can I counter it with "prevent all combat damage" or "counter target spell?"

Mana source is irrelevant now, and does not count as a land. Only "land" types can be destroyed by "destroy land" cards. Preventing combat damage will not do anything against destroy effects, as they do not inflict any damage at all. They simply destroy. Counter spells will work on any spell, regardless off effect (unless it says "this card cannot be countered").

It means spells can't be placed above it on the stack.

Instant. It's yet another term that was taken out, to try and avoid confusion.
